Genotoxicity: Temozolomide has mutagenic effect on bacteria in vitro (Ames test) and fission effect on mammalian cell chromosomes (human peripheral blood lymphocyte test). Reproductive toxicity: No reproductive toxicity studies have been conducted on temozolomide, but repeated toxicity studies in rats and dogs have shown that temozolomide is toxic to animal testes at doses of 50mg/m2 and 125mg/m2 in rats and dogs (approximately 1/4 and 5/8 of the maximum recommended daily human dose based on body surface area, respectively). The appearance of syncytial cells (i.e. immature sperm) and atrophy of testis.

Carcinogenicity: Temozolomide has not been routinely tested for cancer. Temozolomide 125mg/m2 (based on body surface area and equivalent to the maximum recommended daily dose for humans) was administered to rats every 28 days for 5 consecutive days, and breast cancer was developed in both male and female rats after 3 cycles of administration. After 6 cycles of 25, 50, 125mg/m2(approximately 1/8 to 1/2 of the maximum recommended daily human dose based on body surface area), breast cancer occurred in all dose groups: In the high-dose group, fibrosarcomas were found in the tissues of heart, eye, seminal vesicle, salivary gland, abdominal cavity, uterus and prostate, as well as seminal vesicle carcinoma, cardiac schwannoma, optic nerve carcinoma and Hadger's adenocarcinoma. Adenomas were also found in the tissues of skin, lung, pituitary and thyroid.
